           Crystal Violet/LCV ELISA Test Kit (patent-pending) is designed for routine screening for Crystal Violet/Leuco crystal Violet in fish, shrimp, fish/shrimp feed, fish oil and water samples of fish farms or fishponds .The unique features of the kit are:

• High recovery (75 - 115%), and rapid extraction method for fish/shrimp or water samples.
• Converting leucocrystal Violet (LCV) to Crystal Violet before detection. Chromatic Crystal Violet can be rapidly bioconverted into colorless LCV in fish (~80%). The special Oxidant Solution in the kit can convert LCV back to the Crystal Violet form, which can be detected by the kit.
• Providing a 96-well high throughput quantitative detection method. The detection limit is 0.2 ng/g or ppb for fish/shrimp samples.
• The prepared samples can be directly used for liquid chromatography confirmation.
• High reproducibility.

Procedure Overview

The method is based on a competitive colorimetric ELISA assay. The Crystal Violet (CV) has been coated in the plate wells. During the analysis, sample extract and calibrators is added into the wells followed by the CV-antibody to initiate the reaction.After this 30 minute incubation the wells are washed with laboratory grade water. Add CV-HRP conjugate for 30 minutes. Following a second wash after the incubation, a clear substrate is added to the wells and any bound enzyme conjugate causes the conversion to a blue color. After the 15 minute incubation, the reaction is stopped and the amount of color in each well is read . The color of the unknown samples is compared is compared to the color of the calibrators and the CV- concentration of the samples is derived.

Kit Contents, Storage and Shelf Life

            Crystal Violet/LCV ELISA Test Kit has the capacity for 96 determinations or testing of 42 samples in duplicate (assuming 12 wells for standards). The kit components should be stored at 2-8°C*. Return any unused microwells to the foil bag and reseal them with the desiccant provided in the original package. The shelf life is 12 months when the kit is properly stored.
